Functional characterization of insect Chemosensory Proteins (CSPs)


We have described a novel family of proteins, the Chemosensory Proteins (CSPs), which may mediate contact chemosensing in insects. The originality of CSPs is that they may play additional roles in the insect physiological system. They are found everywhere in the insect body at all stages of the insect development, suggesting a dual function sensing/development for this protein family.
We have characterized the 3D structure of the silkworm moth Bombyx mori CSP1. The structure of BmorCSP1 makes up a prism (six alpha-helices) that has capacity to adopt a unique conformational change. This mechanism typical to CSP probably underlies the specific function.


Completing our current investigations on CSPs, the proposed research project addresses the identification of molecular elements underlying CSP function using moths and whiteflies as key models. The key objectives are to:

  • identify the cognate ligands of CSPs and in particular of BmorCSP1 and BtabCSP1,
  • determine the functional structure of the ligand-bounded proteins. Particular attention will be then given to the regulation of the flexible structure of CSP. The expected result is solved function of this new class of protein.


The various tasks to be carried out in this project fall into a multidisciplinary field of modern experimental technologies that include gene cloning/molecular biology and protein expression/purification coupled to binding and structural studies (spectroscopy, NMR, crystallization; collaboration with national and international institutes). RNA interference experiments may be also performed to assess further the CSP functions.
4.Experience in molecular biology, protein biochemistry and biophysics

  • Molecular biology:RNA extraction, cDNA synthesis, Polymerase Chain Reaction (PCR), cloning and molecular sequencing, RNAi
  • Biochemistry:Bacterial expression and protein purification, protein analysis (electrophoresis, western blot, micro-sequencing, MALDI) and binding assay.
  • Proteomics and Biophysics:Spectroscopy and physical chemistry, analysis of protein structures and molecular interactions, supra-molecular assembly, NMR, crystallogenesis, crystallography.
